DISEASE: Defects in BDNF are a cause of congenital central hypoventilation syndrome (CCHS); also known as congenital failure of autonomic control or Ondine curse. CCHS is a rare disorder characterized by abnormal control of respiration in the absence of neuromuscular or lung disease, or an identifiable brain stem lesion. A deficiency in autonomic control of respiration results in inadequate or negligible ventilatory and arousal responses to hypercapnia and hypoxemia. Cross Reactivity:
No cross-reactivity is observed for nerve growth factor (NGF), neurotrophin-3 (NT-3), NT-4/5, glial cell line-derived neurotrophic factor (GDNF) and vascular endothelial growth factor (VEGF165) tested at 25 ng/mL in assay buffer. The reactivity of full-length proBDNF (0.125 ng/mL - 5 ng/mL) was determined in six independent assays using proBDNF proteins from four different sources (mammalian and bacterial, wild-type and mutated). The average cross-reactivity of proBDNF was found to be 5.3% +/- 0.5% in weight (w/v) concentration, or 12.1% +/- 1.2% in molar concentration (mean +/- SEM). The Biosensis proBDNF Rapid TM enzyme-linked immunosorbent assay (ELISA) Kit is a sandwich ELISA that allows the specific, fast and reliable quantification of proBDNF in less than 4 hours in cell culture supernatants, human serum and EDTA-plasma only if used as directed. Please refer to the kit protocol for specific use instructions for each substrate application, in particular human blood samples. This ELISA kit consists of a pre-coated monoclonal anti-proBDNF capture antibody, a biotinylated anti-matureBDNF detection antibody and horseradish peroxidase (HRP)-conjugated streptavidin. The addition of a substrate (3,3',5,5'-tetramethylbenzidine, TMB) yields a colored reaction product which is directly proportional to the concentration of proBDNF present in samples and protein standards. A proBDNF positive control (QC sample) is provided to assure consistent assay performance. This proBDNF ELISA kit employs a recombinant, cleavage-resistant human proBDNF standard produced in mammalian cells by Biosensis and validated against externally available proBDNF proteins. Note that accurate proBDNF quantification in human serum and EDTA-plasma requires the addition of Heterophilic Antibody Blocker BL-004-500 provided in the kit, and available for purchase separately . Specificity:
Human proBDNF. The capture antibody used in this ELISA kit binds to an epitope within the pro-domain of proBDNF. Thus, this ELISA detects the full length form of proBDNF and does not quantify mature BDNF. Whether this ELISA kit can detect truncated BDNF is unknown at present. Cytokeratins 5, 6 and 18 are reported to be expressed in a broad range of human epithelial tissues, from simple glandular epithelia to stratified squamous epithelia. These include epithelial cells that are ectodermal, mesodermal, or endodermal in origin. These cytokeratins have been reported to be expressed in tumor cells of epithelial origin and less commonly of mesothelial origin. Non-epithelial tumors such as lymphomas do not express these cytokeratins. The recognition of cytokeratin 18 on formalin fixed paraffin embedded sections using clone LP34 may be variable.

The 5-HT7 Receptor Antibody was raised against synthetic peptide sequence corresponding to amino acids 8-23 of the rat 5-HT7 receptor coupled to carrier protein with glutaraldehyde. The ImmunoStar 5-HT7 receptor antiserum was quality control tested using standard immunohistochemical methods. The antiserum demonstrates strongly positive labeling of rat cortex and hippocampus using indirect immunofluorescent and biotin/avidin-HRP techniques. Recommended primary dilutions are 1/100 - 1/300 in PBS - biotin/avidin-HRP Technique. Note: use of Triton X-100 or other detergents is not recommended. The antibody was characterized by immunohistochemistry. Immunohistochemical staining of rat brain correlates well with Northern blot analysis, in situ hybridization and receptor autoradiography studies. Immunolabeling is completely abolished by preadsorption with synthetic rat 5-HT7 receptor (8-23). BlastP database sequence homology searches indicate that the amino acid sequence is unique to rat 5-HT7A, 5-HT7B and 5-HT7C. There is also significant sequence overlap with the mouse and human forms.
The 5-HT 2A Receptor Antibody was raised against a multiple antigenic peptide of an N-terminal synthetic sequence corresponding to amino acids 22-41 of rat 5HT2A receptor. The antibody is provided as 100 uL of affinity purified serum in PBS (0.02 M sodium phosphate with 0.15 M sodium chloride, pH 7.5) with 1% BSA (bovine serum albumin), and 0.02% sodium azide. The antiserum demonstrates strongly positive labeling of rat cortex, amygdala and hippocampus using indirect immunofluorescent and biotin/avidin-HRP techniques. Recommended primary dilutions are 1/300 - 1/500 in PBS/0.03% Triton X-100 - Bn/Av-HRP Technique. The addition of intensifying reagents such as nickel ammonium sulfate to the chromogen solution will approximately double the dilution factor as recommended. Immunolabeling is completely abolished by preadsorption with synthetic rat 5HT2A receptor (22-41). Immunolabeling of Western blot revealed a single band of approximately 53kD.
The antibody reacts with the 4th and 5th fibronectin-like repeats of human tenascin-C. It has been demonstrated that tenascin immunoreactivity in breast carcinoma cells could be indicative of metastasis and survival. Recent studies using retrospective material showed that the expression of tenascin in invasion border of early breast cancer significantly correlates with higher risk of distant metastasis. These studies have been continued now and the preliminary results clearly suggest that expression of tenascin in invasion border of early breast cancer is significantly associated with proliferative activity and higher risk of local recurrence. This result implicates a wide application for tenascin antibodies in the breast pathology.

PCNA (proliferating cell nuclear antigen) is the DNA polymerase delta auxiliary protein acting in homotrimeric form to increase the processivity of leading strand synthesis during DNA replication. PCNA is expressed in the nucleus of all proliferating cells. In response to DNA damage, it is ubiquitinated and is involved in the RAD6-dependent DNA repair pathway. PCNA is a useful marker of DNA synthesis, as its form not involved in DNA synthesis degradates in histological preparations in the presence of organic solvents.
