The protein encoded by this gene is a member of the PP2C family of Ser/Thr protein phosphatases. PP2C family members are known to be negative regulators of cell stress response pathways. This phosphatase dephosphorylates, and negatively regulates the activities of, MAP kinases and MAP kinase kinases. It has been shown to inhibit the activation of p38 and JNK kinase cascades induced by environmental stresses. This phosphatase can also dephosphorylate cyclin-dependent kinases, and thus may be involved in cell cycle control. Overexpression of this phosphatase is reported to activate the expression of the tumor suppressor gene TP53/p53, which leads to G2/M cell cycle arrest and apoptosis. Three alternatively spliced transcript variants encoding distinct isoforms have been described.

Pirh 2 (P53 induced RING-H2 protein), also known as RCHY1, it forms dimers through its N- and C-terminus in cells. The Pirh2 has ubiquitin-protein ligase activity and it binds with p53 and promotes the ubiquitin-mediated proteosomal degradation of p53. The Pirh2 is oncogenic because loss of p53 function contributes directly to malignant tumor development. Pirh2 expression decreases the level of p53, and a decrease of endogenous Pirh2 expression increases p53 levels. Pirh2 is therefore considered, together with MDM2, to act as a negative regulator of p53 function.

Lyn (also known as p53/56 Lyn) is a membrane-associated protein tyrosine kinase (PTK) mostly expressed in hemopoietic cells which is important in cellular signaling. It contains an SH2 and SH3 domain and has been found to be cleaved after activation of caspases in apoptosis. A member of the Src family of PTKs, there are two known isoforms for Lyn which plays an indispensable role in the Fc epsilon RI (Fcer1) and the B-cell IgM receptor signaling pathway and is essential for Syk activation and Lat phosphorylation after Fcer1 aggregation and can also phosphor-ylate Tec on multiple residues. Lyn can also be regulated by IL-2 and IL-3.Lyn is a member of the src family of non-receptor protein tyrosine kinases that is predominantly expressed in haematopoietic tissues. Like all members of the src family, lyn is thought to participate in signal transduction from cell surface receptors that lack intrinsic tyrosine kinase activity. It is associated with a number of cell surface receptors including the B cell antigen receptor and immunoglobulin E receptor (FceRI).

CHK2: CHK2 checkpoint homolog (S. pombe). In response to DNA damage and replication blocks, cell cycle progression is halted through the control of critical cell cycle regulators. The protein encoded by this gene is a cell cycle checkpoint regulator and putative tumor suppressor. It contains a forkhead-associated protein interaction domain essential for activation in response to DNA damage and is rapidly phosphorylated in response to replication blocks and DNA damage. When activated, the encoded protein is known to inhibit CDC25C phosphatase, preventing entry into mitosis, and has been shown to stabilize the tumor suppressor protein p53, leading to cell cycle arrest in G1. In addition, this protein interacts with and phosphorylates BRCA1, allowing BRCA1 to restore survival after DNA damage. Mutations in this gene have been linked with Li-Fraumeni syndrome, a highly penetrant familial cancer phenotype usually associated with inherited mutations in TP53. Also, mutations in this gene are thought to confer a predisposition to sarcomas, breast cancer, and brain tumors. This nuclear protein is a member of the CDS1 subfamily of serine/threonine protein kinases. Three transcript variants encoding different isoforms have been found for this gene.

CHK1, also known as CHEK1, it is a kinase that phosphorylates cdc25, an important phosphatase in cell cycle control, particularly for entry into mitosis. Cdc25, when phosphorylated on serine 216 by chk1 becomes bound by an adaptor protein in the cytoplasm. Therefore it is inhibited from removing the inhibiting phosphate from MPF (mitotic/maturation promoting factor) added by Wee1. Consequently, a cell is prevented from entering mitosis. Chk1 can also phosphorylate p53 at Ser20 in vitro.

This gene generates several transcript variants which differ in their first exons. At least three alternatively spliced variants encoding distinct proteins have been reported, two of which encode structurally related isoforms known to function as inhibitors of CDK4 kinase. The remaining transcript includes an alternate first exon located 20 Kb upstream of the remainder of the gene; this transcript contains an alternate open reading frame (ARF) that specifies a protein which is structurally unrelated to the products of the other variants. This ARF product functions as a stabilizer of the tumor suppressor protein p53 as it can interact with, and sequester, the E3 ubiquitin-protein ligase MDM2, a protein responsible for the degradation of p53. In spite of the structural and functional differences, the CDK inhibitor isoforms and the ARF product encoded by this gene, through the regulatory roles of CDK4 and p53 in cell cycle G1 progression, share a common functionality in cell cycle G1 control. This gene is frequently mutated or deleted in a wide variety of tumors, and is known to be an important tumor suppressor gene.

C17orf53 (chromosome 17 open reading frame 53) is a 647 amino acid protein that is encoded by a gene mapping to human chromosome 17. Chromosome 17 makes up over 2.5% of the human genome with about 81 million bases encoding over 1,200 genes. Two key tumor suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. Tumor suppressor p53 is necessary for maintenance of cellular genetic integrity by moderating cell fate through DNA repair versus cell death. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome. Like p53, BRCA1 is directly involved in DNA repair, specifically it is recognized as a genetic determinant of early onset breast cancer and predisposition to cancers of the ovary, colon, prostate gland and fallopian tubes. Chromosome 17 is also linked to neurofibromatosis, a condition characterized by neural and epidermal lesions, and dysregulated Schwann cell growth. Alexander disease, Birt-Hogg-Dube syndrome and Canavan disease are also associated with chromosome 17. ; ;

The tumour suppressor protein p53 is a key element of intracellular anticancer protection. It mediates cell cycle arrest or apoptosis in response to DNA damage or to starvation for pyrimidine nukleotides. It is up-regulated in response to these stress signals and stimulated to activate transcription of specific genes, resulting in expression of p21waf1 and other proteins involved in G1 or G2/M arrest, or proteins that trigger apoptosis, such as Bcl-2. The structure of p53 comprises N-terminal transactivation domain, central DNA-binding domain, oligomerisation domain, and C-terminal regulatory domain. There are various phosphorylation sites on p53, of which the phosphorylation at Ser15 is important for p53 activation and stabilization.

Ku Antigen (DNA-dependent DNA helicase) is a heterodimer (of 72 and 87 kDa polypeptides) which contributes to genomic integrity through its ability to bind DNA double-strand breaks and facilitate repair by the nonhomologous end-joining pathway. A DNA double-strand break is a major lesion that destroys the integrity of the DNA molecule. Such damage is introduced by ionizing radiation. Ku binds to free double-stranded DNA ends and is the DNA-binding component of the DNA-dependent protein kinase. Thus, the Ku protein is involved in DNA repair and in V(D)J recombination, and the Ku-DNA-dependent protein kinase complex may have a role in those same processes. Ku70 and Ku80 share a common topology and form a dyad-symmetrical molecule with a preformed ring that encircles duplex DNA. The binding site can cradle 2 full turns of DNA while encircling only the central 3-4 base pairs. Ku makes no contacts with DNA bases and few with the sugar-phosphate backbone, but it fits sterically to major and minor groove contours so as to position the DNA helix in a defined path through the protein ring. These features are well designed to structurally support broken DNA ends and to bring the DNA helix into phase across the junction during end processing and ligation. Mouse cells deficient for Ku80 display a marked increase in chromosomal aberrations, including breakage, translocations, and aneuploidy. Despite the observed chromosome instabilities, Ku80 -/- mice have only a slightly earlier onset of cancer. Loss of p53 synergizes with Ku80 to promote tumorigenesis such that all Ku80 -/- and p53 -/- mice succumb to disseminated pro-B-cell lymphoma before 3 months of age. The p70/p80 complex binds to the ends of double-stranded DNA in a cell cycle-dependent manner, being associated with chromosomes of interphase cells, followed by complete dissociation from the condensing chromosomes in early prophase. Some patients with systemic lupus erythematosus produce very large amounts of autoantibodies to p70 and p80. The autoantibody has been found in patients with autoimmune thyroid disease (Graves disease) as well as in those with lupus.

The antibody MEM-54 reacts with Ku80, a 80 kDa subunit of Ku autoantigen (heterodimer of 72 and 87 kDa intracellular polypeptides). Ku autoantigen is involved in DNA repair and in V(D)J recombination through its ability to bind DNA double-strand breaks.
Lyn is a Src-family protein tyrosine kinase that is predominantly expressed in hematopoietic cells. It is associated with a number of cell surface receptors including the B cell antigen receptor (BCR) and Fc receptors. Upon their triggering, Lyn phosphorylates subunits of these receptors in a cholesterol-dependent manner, utilizing the plasma membrane lipid raft system. The phosphorylated intracellular domains of the receptors are accessible for cytoplasmic Syk tyrosine kinase, which is activated by Lyn-mediated phosphorylation and which transduces the signal to downstream adaptors. Lyn is abnormally distributed in acute myeloid leukemia cells and seems to be a novel pharmacologic target of this disease.

The p53 family of proteins includes three members, p53, p63, and p73. The protein p63 is encoded by TP63 gene, which gives rise to protein isoforms with different properties and functions due to the presence (TAp63) or absence (deltaNp63) of an N-terminal transactivation domain. Immunohistochemistry of p63 has a clinical relevance for certain tumor types, but investigations have been hampered by a lack of well characterized antibodies that are specific for p63, do not cross-react with the related p53 and p73 proteins, and allow for discrimination between p63 isoforms TAp63 and deltaNp63 with opposite functional properties.
Cdc25C is a tyrosine phosphatase and belongs to the Cdc25 phosphatase family. It has been highly conserved during evolution and it plays a key role in the regulation of cell division. It directs dephosphorylation of cyclin B-bound CDC2 and triggers entry into mitosis. It is also thought to suppress p53-induced growth arrest. Cdc25C is mainly expressed in G2 phase. Multiple alternatively spliced transcript variants of this gene have been described, however, the full-length nature of many of them is not known.
