CD58 (LFA-3) is an immunoglobulin family adhession molecule expressed by both hematopoietic and non-hematopoietic cells (often on antigen presenting cells) and serving as ligand of CD2. This interaction is important for T cell-mediated immunity. CD58 is expressed in transmembrane form and in GPI-anchored form; the later is constitutively associated with protein kinases whereas the transmembrane form activates kinase activity upon triggering. CD58 is a powerful tool for detection of minimal residual disease in acute lymphocytic leukemia, and for evaluation of liver damage related with hepatitis B.

CD20 is a cell surface 33-37 (depending on the degree of phosphorylation) kDa non-glycosylated surface phosphoprotein expressed on mature and most malignant B cells, but not stem cells or plasma cells (low number of the CD20 has been also detected on a subpopulation of T lymphocytes and it can be expressed on follicular dendritic cells). Its expression on B cells is synchronous with the expression of surface IgM. CD20 regulates transmembrane calcium conductance (probably functioning as a component of store-operated calcium channel), cell cycle progression and B-cell proliferation. It is associated with lipid rafts, but the intensity of this association depends on extracellular triggering, employing CD20 conformational change and/or BCR (B cell antigen receptor) aggregation. After the receptor ligation, BCR and CD20 colocalize and then rapidly dissociate before BCR endocytosis, whereas CD20 remains at the cell surface. CD20 serves as a useful target for antibody-mediated therapeutic depletion of B cells, as it is expressed at high levels on most B-cell malignancies, but does not become internalized or shed from the plasma membrane following mAb treatment.

This gene encodes a chromatin-associated enzyme, poly(ADP-ribosyl)transferase, which modifies various nuclear proteins by poly(ADP-ribosyl)ation. The modification is dependent on DNA and is involved in the regulation of various important cellular processes such as differentiation, proliferation, and tumor transformation and also in the regulation of the molecular events involved in the recovery of cell from DNA damage. In addition, this enzyme may be the site of mutation in Fanconi anemia, and may participate in the pathophysiology of type I diabetes.

PTK6 (protein tyrosine kinase 6, BRK or FLJ42088), with 451-amino acid protein (about 52kDa), encods a cytoplasmic nonreceptor protein kinase which may function as an intracellular signal transducer in epithelial tissues. Its presence in the nucleus appears to be linked to suppression of tumor progression. The encoded protein has been shown to undergo autophosphory-lation. Very high level in colon and high levels in small intestine and prostate, and low levels in some fetal tissues. And Expressed at low level in some breast tumors, but not in normal breast. Also found in melanocytes, but not expressed in heart, brain, placenta, lung, liver, skeletal muscle, kidney and pancreas. Overexpression of this gene in mammary epithelial cells leads to sensitization of the cells to epidermal growth factor and results in a partially transformed phenotype.

The nuclear lamina consists of a two-dimensional matrix of proteins located next to the inner nuclear membrane. The lamin family of proteins make up the matrix and are highly conserved in evolution. During mitosis, the lamina matrix is reversibly disassembled as the lamin proteins are phosphorylated. Lamin proteins are thought to be involved in nuclear stability, chromatin structure and gene expression. Vertebrate lamins consist of two types, A and B. Through alternate splicing, this gene encodes three type A lamin isoforms. Mutations in this gene lead to several diseases: Emery-Dreifuss muscular dystrophy, familial partial lipodystrophy, limb girdle muscular dystrophy, dilated cardiomyopathy, Charcot-Marie-Tooth disease, and Hutchinson-Gilford progeria syndrome.

CD13 antigen, also known as aminopeptidase N, is a member of type II integral membrane metalloproteases, which also includes the leukocyte antigens CD10, CD26, CD73 and BP-1. CD13 antigen is a receptor for the coronaviruses which cause respiratory disease in humans and several animal species. The antigen functions as a zinc-binding metalloprotease which plays a role in cell surface antigen presentation by trimming the N-terminal amino acids from MHC class II-bound peptides. CD13 antigen is reported to be expressed on granulocytes, monocytes and their precursors, most acute myeloid leukemias and a smaller proportion of acute lymphoid leukemias. Non-hematopoietic cells which express CD13 antigen include epithelial cells, renal proximal tubules, intestinal brush border, endothelial cells, fibroblasts, brain cells, bone marrow, osteoclasts and cells lining the bile canaliculi.

The Mu Opioid Receptor antiserum was quality control tested using standard immunohistochemical methods. The antiserum demonstrates strongly positive labeling of rat caudate putamen and spinal cord (dorsal horn) using indirect immunofluorescent and biotin/avidin-HRP techniques. Recommended primary dilutions are 1/500 - 1/1000 in PBS/0.3% Triton X-100 - Cy3 Technique and 1/6000 - 1/10000 in PBS/0.3% Triton X-100 - Biotin/avidin-HRP Technique. Preadsorption with MOR peptide (384-398) at 10 µg/ml completely eliminates labeling. The specificity of the antiserum was determined by immunolabeling of transfected cells, Western Blot analysis and immunoisolation studies.

Amylo-Glo RTD Ready to Dilute Staining reagent is designed to stain amyloid plaques in tissue sections. This novel marker has several advantages over other conventional markers such as Thioflavin S and Congo Red because of its unique chemical and spectral properties. (L. Schmued et al. (2012) J.Neuroscience Methods 209:120- 126). Using Amylo-Glo results in a very bright blue UV excitable stain under physiological conditions that will not bleed through when illuminated with other filters. Its brightness makes it ideal for low magnification quantification studies, while its unique excitation/emission profile and mild staining conditions makes it ideal for combination for multiple immunofluorescent labeling studies. Amylo-Glo RTD is compatible with fresh, frozen, and formalin-fixed immunohistochemistry or cytochemistry, and it is particularly good for confocal and multiple labeling because of its high fluorescent intensity and high resistance to photo-bleaching. Moreover because Amylo-Glo fluoresces in the UV channel, double and triple labeling experiments can be performed very easily (see protocol).

After reconstitution keep aliquots at -20°C for a higher stability, and at 2-8°C with an appropriate antibacterial agent. Avoid repetitive freeze/thaw cycles. Glycerol (1:1) may be added for an additional stability.
Rabbit anti-Autophagy-related protein 16-1 (ATG16 L1) Polyclonal Antibody (Unconjugated), suitable for WB, ELISA, ICC.
Background Info:
ATG16 L1 plays an essential role in autophagy. It is a homooligomer ant interacts with ATG5. Part of either the minor and major complexes respectively composed of 4 sets of ATG12-ATG5 and ATG16L1 (400 kDa) or 8 sets of ATG12-ATG5 and ATG16L1 (800 kDa). APG16 L1 localises to the cytoplasm, in particularly to preautophagosomal structure (PAS) where it is involved in the membrane targeting of ATG5. There are 5 named isoforms produced by alternative splicing. Genetic variation within ATG16 L1 are associated with susceptibility to inflammatory bowel disease type 1.

Rabbit anti-Gamma-synuclein Polyclonal Antibody (Unconjugated), suitable for WB, IHC-Frozen.
Background Info:
FUNCTION: Plays a role in neurofilament network integrity. May be involved in modulating axonal architecture during development and in the adult. In vitro, increases the susceptibility of neurofilament-H to calcium-dependent proteases. May also function in modulating the keratin network in skin. Activates the MAPK and Elk-1 signal transduction pathway. SUBUNIT: May be a centrosome-associated protein. SUBCELLULAR LOCATION: Cytoplasm; perinuclear region. Centrosome. Spindle. Associated with centrosomes in several interphase cells. In mitotic cells, localized to the poles of the spindle. TISSUE SPECIFICITY: Highly expressed in brain, particularly in the substantia nigra. Also expressed in the corpus callosum, heart, skeletal muscle, ovary, testis, colon and spleen. Weak expression in pancreas, kidney and lung. PTM: Phosphorylated. Phosphorylation by GRK5 appears to occur on residues distinct from the residue phosphorylated by other kinases. DISEASE: Brain iron accumulation type 1 (NBIA1, also called Hallervorden-Spatz syndrome), a rare neuroaxonal dystrophy, is histologically characterized by axonal spheroids, iron deposition, Lewy body (LB)-like intraneuronal inclusions, glial inclusions and neurofibrillary tangles. SNCG is found in spheroids but not in inclusions. SIMILARITY: Belongs to the synuclein family.
