Glial fibrillary acidic protein (GFAP) is an intermediate filament protein of 52kD reported to be expressed in glial cells, for example, astrocytes and ependymal cells. In the peripheral nervous system, GFAP has been reported to be expressed in Schwann cells, enteric glial cells and satellite cells of human sensory ganglia and in neoplastic tissues GFAP has been reported to be expressed in astrocytomas and ependymomas. When using GFAP-GA5 the heat induced epitope retrieval (HIER) technique may improve staining in some cases.

Human fascin is a 55 to 58 kD actin-bundling protein, whose actin binding ability is regulated by phosphorylation. In normal tissues the detection of fascin is reported to be predominantly restricted to dendritic cells, and in the thymus has been observed only in medullary dendritic cells. In reactive nodes, interdigitating reticulum cells of T cell zones, cells in subcapsular areas, and cells of the reticular network express fascin. Variable expression is seen in follicular dendritic cells and endothelial cells. Lymphoid cells, myeloid cells and plasma cells do not express fascin; however, in cases of Hodgkin's disease, including nodular sclerosis, mixed cellularity lymphocyte depletion and unclassified cases, most or all Reed Sternberg cells are reported to be positive for fascin. Fascin expression may be induced by Epstein-Barr virus (EBV) infection of B cells with the possibility that viral induction of fascin in lymphoid or other cell types must also be considered in EBV-positive cases.

Terminal deoxynucleotidyl transferase (TdT) is a DNA polymerase of 58 kD located in the cell nucleus which catalyzes the polymerization of deoxynucleotides at the 3' hydroxyl ends of oligo or polydeoxynucleotide initiators and functions without a template. TdT is reported to be expressed in primitive T and B lymphocytes of the normal thymus and bone marrow. The identification of TdT-positive cell populations in primary and secondary lymphoid organs during maturation of the immune system is one area of interest but it is the reported occurrence of high levels of enzyme activity in white blood cells and bone marrow in certain leukemias which is of particular interest.

Nectin (originally isolated as poliovirus receptor-related protein (PRR)) is a cell-cell adhesion molecule of the immunoglobulin supergene family. Nectin is colocalized with afadin at cadherin-based cell-cell adherence junctions in various tissues and cell lines. The nectin family consist of nectin-1, nectin-2 and nectin-3. Nectin-2 also known as PRR2 or CD112 is a plasma membrane adhesion molecule localized at adherens junctions which is widely expressed in various cell lines including neuronal, endothelial, epithelial and hematopoietic cells. Next to its role in adherens junctions it functions as alphaherpus virus receptor and acts also as intercellular adhesion molecule and pseudorabies virus receptor. Disruption of mouse nectin-2 leads to infertility of male mice. Monoclonal antibody 502-57 has been raised against the extracellular domain of mouse nectin-2. DAPI (4',6-diamidino-2-phenylindole) is a fluorescent dye which can bind DNA strands robustly, the fluorescence being detected by fluorescence microscope. DAPI can dye both live and fixed cells as it can cross intact membrane, with higher efficiency in fixed cells. The molecular formula is C16H15N5·2HCl with 350.25 g/mol molecular weight, CAS Number 28718-90-3. DAPI could pass through the cell and nucleic membranes and bind the double-strand DNA in the nucleus, producing 20 times stronger fluorescence than itself. The efficiency detected by fluorescence microscope is very high (almost 100%), having no side effects for the live cells. The sensitivity for double stranded DNA DAPI staining is many times larger comparing to ethidium bromide (EB). DAPI staining is usually used in cell death detection, as it enters more effectively and generates stronger fluorescence in dead cells. After staining with DAPI, detect with fluorescence microscope or flow cytometry. Blue fluorescent cell would be seen under the microscope after staining. The largest excitation wavelength for DAPI is 340nm (ultraviolet), and the largest emission wavelength is 488nm (blue). When DAPI binds with double-strand DNA, the largest excitation wavelength is 360nm, while the largest emission wavelength becomes 460nm. DAPI's blue emission makes it suitable for combined assays where the fluorescence ranges of DAPI and other IHC-employed fluorescent molecules like green-fluorescent fluorescein and GFP, or red-fluorescent stains like Texas Red, are completely distinctive.

The VMAT1 antiserum was quality control tested using standard immunohistochemical methods in rat adrenal medulla using biotin/avidin-HRP techniques; the antiserum shows no reactivity in rat CNS. Specificity of the antiserum was demonstrated by soluble pre-adsorption and Western blot. Tissue staining is completely eliminated by pretreatment of the diluted antibody with an excess of rat VMAT1 peptide residues 502-521. Western blot analysis of immunoprecipitated rat adrenal homogenates demonstrates a dense immunoreactive band of approximately 55 kD and a minor band of approximately 75 kD.
