Anti-p53 (Clones PAb1620 and PAb122)

 

p53 acts as a tumor suppressor in many tumor types; depending on the physiological circumstances and cell type it induces growth arrest or apoptosis. Involved in cell cycle regulation as a trans-activator, p53 acts to negatively regulate cell division by controlling a set of genes required for this process. Apoptosis induction seems to be mediated either by stimulation of BAX and FAS antigen expression, or by repression of Bcl-2 expression.

 

p53 localizes in the nucleus, yet can be chaperoned to the cytoplasm by the negative regulator, MDM2. p53 fluctuates between latent and active DNA-binding conformations and is differentially activated through post-translational modifications, including phosphorylation and acetylation.

Anti p53 Clone PAb122 binds to amino acids 371 - 378 of p53 and is recommended for the detection of a conserved denaturation-resistant determinant of the p53 protein.

 

Anti p53 Clone PAb1620 binds a conformational epitope that disappears in some mutant p53 or upon denaturation. It therefore preferentially immunoprecipitates wild-type p53 and does not cause immunoprecipitation of mutant and denatured p53.
