Saposins A-D localize primarily to the lysosomal compartment where they facilitate the catabolism of glycosphingolipids with short oligosaccharide groups. The precursor protein exists both as a secretory protein and as an integral membrane protein and has neurotrophic activities. Daxx is an apoptosis-modulating death domain-associated protein with functions in transcriptional regulation. Daxx functions both in cytoplasm, where it interacts with Fas, and in nucleus (residing in PML oncogenic domains), where it is involved in SUMO-dependent regulation of transcription and subnuclear compartmentalization. A point mutation in the gene coding for the alpha-synuclein protein was the first discovery linking this protein to a rare familial form of Parkinson's disease (PD). Subsequently, other mutations in the alpha-synuclein gene have been identified in familial PD. The aggregated proteinaceous inclusions called Lewy bodies found in PD and cortical Lewy body dementia (LBD) were discovered to be predominantly alpha-synuclein. Aberrant aggregation of alpha-synuclein has been detected in an increasing number of neurodegenerative diseases, collectively known as synucleopathies. Alpha-synuclein exists physiologically in both soluble and membrane-bound states, in unstructured and alpha-helical conformations, respectively. The physiological function of alpha-synuclein appears to require its translocation between these subcellular compartments and interconversion between the 2 conformations. Abnormal processing of alpha-synuclein is predicted to lead to pathological changes in its binding properties and function.

The monoclonal antibody 5G5 reacts with the Toll-like receptor 9 (TLR9, CD289). TLRs are highly conserved throughout evolution and have been implicated in the innate defence to many pathogens. In Drosophila, toll is required for the anti-fungal response, while the related 18-wheeler is involved in antibacterial defences. In mammals, TLRs identified as type I transmembrane signalling receptors with pattern recognition capabilities, have been implicated in the innate host defence to pathogens. As investigated so far all functional characterized TLR signal via the TLR/IL-1 receptor (IL-1R) pathway where recruitment of MyD88 seems to be essential. Desmoglein 3 is a member of the desmosomal cadherin family that comprise the desmogleins and desmocollins. This ELISA kit consists of a pre-coated sheep polyclonal anti-alpha-synuclein (aa: 116-131) capture antibody, a biotinylated mouse monoclonal anti-alpha-synuclein detection antibody (aa: 61-95) and horseradish peroxidase (HRP)-conjugated streptavidin. The addition of a substrate (3,3',5,5'-tetramethylbenzidine, TMB) yields a colored reaction product which is directly proportional to the concentration of alpha-synuclein present in samples and protein standards. A human alpha-synuclein positive control (QC sample) is provided to assure consistent assay performance. This ELISA kit has not been tested for other substrate applications, only citrate plasma and CSF, other substrates have not been tested and performance may vary. Background Info:
Alpha synuclein is an abundant 140 amino acid neuronal protein, expressed primarily at presynaptic terminals in the central nervous system. Alpha synuclein has been associated with several neurodegenerative diseases. A point mutation in the gene coding for the alpha-synuclein protein was the first discovery linking this protein to a rare familial form of Parkinson's disease (PD). Subsequently, other mutations in the alpha-synuclein gene have been identified in familial PD. The aggregated proteinaceous inclusions called Lewy bodies found in PD and cortical Lewy body dementia (LBD) were discovered to be predominantly alpha-synuclein. Aberrant aggregation of alpha-synuclein has been detected in an increasing number of neurodegenerative diseases, collectively known as synucleopathies. Alpha-synuclein exists physiologically in both soluble and membrane-bound states, in unstructured and alpha-helical conformations, respectively. The physiological function of alpha-synuclein appears to require its translocation between these subcellular compartments and interconversion between the 2 conformations. Abnormal processing of alpha-synuclein is predicted to lead to pathological changes in its binding properties and function.

Cross Reactivity:

Cross-reactivity was calculated based on increase or decrease of apparent ?-synuclein concentrations in spiked samples. The Glycine decarboxylase complex (GDC) is abundant in mitochondria matrix of C3 leaves and functions in photorespiratory carbon recovery. GDC enzyme can account for up to 50% of matrix protein, and is responsible for the most prominent metabolic activity in the mitochondria of illuminated leaves, photorespiration. The dry, unopened container should be stored at room temperature in a dry or desiccated location protected from light. Do not store in the refrigerator unless material is in a dry, moisture free environment. Material is hydroscopic so once the seal is broken it should be hydrated and not resealed while dry. Fibrillarin is part of the small subunit processome complex, involved in the processing of pre-18S ribosomal RNA. Nop1p is the yeast homologue of fibrillarin. Fibrillarin/Nop1p is extraordinarily conserved, so that the yeast and human proteins are 67% identical, and the human protein can functionally replace the yeast protein. This means that suitably cross-reactive antibodies to Nop1p/fibrillarin such as this antibody can be used to reveal nucleoli and study fibrillarin/Nop1p in all eukaryotes and archea tested to date. This antibody is becoming widely used as a convenient marker for nucleoli in a wide variety of species.

A point mutation in the gene coding for the alpha-synuclein protein was the first discovery linking this protein to a rare familial form of Parkinson's disease (PD). Subsequently, other mutations in the alpha-synuclein gene have been identified in familial PD. The aggregated proteinaceous inclusions called Lewy bodies found in PD and cortical Lewy body dementia (LBD) were discovered to be predominantly alpha-synuclein. Aberrant aggregation of alpha-synuclein has been detected in an increasing number of neurodegenerative diseases, collectively known as synucleopathies. Alpha-synuclein exists physiologically in both soluble and membrane-bound states, in unstructured and alpha-helical conformations, respectively. The physiological function of alpha-synuclein appears to require its translocation between these subcellular compartments and interconversion between the 2 conformations. Abnormal processing of alpha-synuclein is predicted to lead to pathological changes in its binding properties and function.

Sensitivity:
Typical limit of detection (LOD) for alpha-synuclein is < 100 pg/mL, determined as alpha-synuclein concentration at blank OD plus 3x standard deviation of blank OD (n=10).
Cross Reactivity:
Interference and cross-reactivity of human beta- and gamma-synuclein was assessed by spiking each protein at excess concentration of 20 ng/mL into two human plasma samples, each diluted 1:10 and 1:20, in comparison to unspiked samples. Cross-reactivity was calculated based on increase or decrease of apparent ?-synuclein concentrations in spiked samples. At a 3-6 fold excess (w/v) of spiked protein over endogenous alpha-synuclein at both sample dilutions, this ELISA shows very little cross-reactivity of 2.3% or less.<br>Individual alpha-synuclein isoforms have not yet been tested, but it is expected that this ELISA kit detects monomeric, oligomeric and phosphorylated alpha-synuclein isoforms.
